Bouin’s Fluid

Bouin’s Fluid is a fixative solution commonly used in histology and pathology to preserve tissue samples for microscopic examination. It is a mixture of picric acid, formaldehyde, and glacial acetic acid. Bouin’s Fluid is known for its excellent tissue preservation qualities, particularly in retaining cellular details and the integrity of soft tissues.

Application:

• Histology: Used as a fixative to preserve tissue samples for microscopic examination, especially for delicate tissues like embryonic tissue, and endocrine organs.
• Immunohistochemistry: Employed in specific staining procedures where it enhances staining quality.
• Morphological Studies: Preferred for tissues where preservation of fine cellular detail is critical.

Handling and Storage:

• Handling:
  • Use in a well-ventilated area or under a fume hood to avoid inhaling fumes.
  • Wear appropriate personal protective equipment (PPE) including gloves, lab coat, and eye protection.
  • Avoid contact with skin and eyes; in case of contact, rinse immediately with plenty of water.
  • Handle with care to avoid spills. In case of a spill, evacuate the area and follow proper spill cleanup procedures.
• Storage:
  • Store in a cool, dry, and well-ventilated area away from incompatible materials like bases and strong oxidizers.
  • Keep container tightly closed when not in use to prevent evaporation and contamination.
  • Store in an approved safety container, clearly labeled, and away from sources of ignition.
  • Monitor for signs of picric acid crystallization, which can lead to explosive hazards. If crystals are observed, do not move the container, and seek professional assistance.

Disposal:

• Dispose of Bouin’s Fluid according to local, regional, or national regulations. Do not pour it down the drain or dispose of it with regular trash. Consider neutralizing formaldehyde and picric acid components before disposal, if feasible.
