Principle

Acetate Differential Agar contains sodium acetate as the primary carbon source. Microorganisms capable of utilizing acetate grow on the medium and produce characteristic reactions, while organisms unable to metabolize acetate exhibit limited or no growth. The resulting growth characteristics assist microbiologists in differentiating and identifying bacterial species during biochemical testing.

Preparation

  1. Suspend the recommended quantity of dehydrated medium in purified water.
  2. Heat with frequent stirring until completely dissolved.
  3. Sterilize by autoclaving at 121°C for 15 minutes.
  4. Cool to 45–50°C and mix thoroughly.
  5. Pour into sterile Petri plates and allow to solidify.

  • Store the dehydrated medium in a tightly closed container.
  • Keep in a cool, dry place away from direct sunlight and moisture.
  • Maintain storage temperature between 10°C and 30°C.
